Biochem/physiol Actions

Spleen associated tyrosine kinase (SYK) is a key component of B cell receptor (BCR) signalling pathway and the disruption of SYK leads to blocking of BCR downstream cell signalling. SYK is also involved in the downstream signalling of T cell receptor (TCR). SYK expression level is high in thymocytes than in mature T cells. SYK is critically required for high-affinity IgE receptor (FcεRI) mediated calcium mobilization, degranulation, cytokine production and metabolism of arachidonic acid. In platelets, SYK through interaction with Fc gamma receptor (FcRγ) is essential for collagen receptor mediated signalling pathway. SYK protects B cells from oxidative stress mediated apoptosis by modulating caspase 9 and activation of serine/threonine-specific protein kinase (Akt) survival pathway. SYK plays a key role in maintaining vascular integrity in vivo. SYK is a tumor suppressor gene in breast cancer. SYK is a prognostic marker for chemonaive lymph node negative colon cancer.

General description

Spleen associated tyrosine kinase (SYK) belongs to an autonomous family of cytosolic non-receptor tyrosine kinases. SYK have a tandem of Src Homology 2 (SH2) domain, a C-terminal kinase domain and lacks SH3 domain. SYK is expressed in neurons, hepatocytes, nasal fibroblast, vascular endothelial cells, hematopoietic cells and mammary epithelial cells. In human chromosome, the gene SYK is localized on 9q22.

Peptide sequence around phosphorylation site of tyrosine 352(S-P-Y(p)-A-D) derived from Human SYK.

Specificity

The antibody detects endogenous levels of SYK only when phosphorylated at tyrosine 352.
